2nd run of AKTA. the result is the same as that of 1st run.

Mon 21th June 2010

Done deoiling and starting of the process of freeze dryng. We are testing on the crude seed (only deoiled). But before freeze drying, we need to freeze the product 1st. So we put the product in at 4:45pm into mr yeo's -20degree C fridge and freeze for 24 hrs. We have 3 small bottles, so we set bottle 1 as 24hrs freezing, bottle 2 as 48hrs freezing, bottle 3 72hrs freezing before we but each to freeze dry for 24hrs.



Purpose of this experiment- see if the duration of freezing will affect the freeze drying quality.

Tues 22th June 2010

Run AKTA with Mr Kon.
get the results above. We did not know why our results not the same as the previous group. Maybe is because our seed put for a long time (26 april till now), so it might degrade.
Or is because the previous group did step elution. this lead to the previous 3 peaks come out together -> therefore 2 peaks.
We are going to try to run a freshly grinded, deoiled protein to see if there is any diff in results.

Wed 23th June 2010

Did not conduct any experiment. Only go to W3110f to change the freeze drying bottle.
Did research and ask Mr Wong on the molecular weight. coz we run out of it for SDS-page..

Thurs 24th June 2010

At last! We run SDS-page for our 1st Akta result.. the photo will be up next mon. not going to do experiment tomorrow. coz Mr Yeo half day. will upoad the pic on mon.

lesson of the day: recap on how to run SDS-page. did encounter some problems like the dilution factors and things like tat. but after looking through mr wong's and mr yeo's notes, and mr yeo's guidance, we complete the SDS-page.

Done water analysis for Mr Ting
Samples from 5 wells in i dun noe where
only 50ml given so we did only phosphate, sulfate, nitrate, ammonia and E.coli tests.

do E.coli test using coliform test, writing water analysis report.

Fri 18th June 2010
FINALLY...we get to run AKTA to get the proteins out of MO seeds.
uses 10.1g of grinded seed powder to deoiled and we run abt 10 to 15 ml into AKTA to get the proteins out.
